Somatic loss of the Y chromosome complements polygenic risk scores for type 2 diabetes risk prediction

Nature Medicine, Published online: 02 March 2026; doi:10.1038/s41591-026-04214-y We assessed how germline Y haplogroups and somatic loss of the Y chromosome (LOY) associate with risk of disease in East Asian and European populations. LOY increases the risk of type 2 diabetes in East Asian men, particularly when polygenic risk scores are low. The identification of pancreatic β-cells with LOY supports its potential involvement in the pathogenesis of type 2 diabetes.
